生物碱试剂本质就是酸类,它们一般均导致蛋白质变性.因为反应机理是 跟蛋白质侧链上阳性离子(如氨基等)发生酸碱成盐反应,而侧链氨基常参与蛋白质的高级结构或酶的活性中心,因此变性.(蛋白质盐是很大的分子,沉淀是很自然的)。
大多数生物碱在酸水或稀醇中能与某些试剂反应生成难溶于水的复盐或分子络合物,这些试剂称为生物碱沉淀试剂.
1.常用的生物碱沉淀试剂
碘化物复盐、重金属盐、大分子酸类等.常见的生物碱沉淀试剂有碘-碘化钾试剂、碘化铋钾试剂、碘化汞钾试剂等.
2.生物碱沉淀反应的条件
(1)反应环境
生物碱沉淀的反应一般在稀酸水溶液中进行.
(2)净化处理
共存的蛋白质、多肽、鞣质等成分,这些物质也能与生物碱沉淀试剂发生沉淀的反应.为了避免其干扰,可将酸水液经碱化后用氯仿萃取,除去水溶性干扰成分,然后用酸水从氯仿中萃取出生物碱,以此酸水液进行沉淀反应.
3.生物碱沉淀反应阳性结果的判断
(1)阳性结果判断
一般需选用三种以上的沉淀试剂进行反应,如果均有生物碱的沉淀反应,可判断为阳性结果.
(2)需要注意的问题
①少数生物碱不能与一般生物碱沉淀试剂产生沉淀反应.如麻黄碱、咖啡碱与多数生物碱沉淀试剂不能发生沉淀反应.
②中药中有些非生物碱类物质也能与生物碱沉淀试剂产生沉淀的应,如蛋白质、多肽、氨基酸、鞣质等.
